Optimal proteome allocation and the temperature dependence of microbial growth laws

چکیده

Abstract Although the effect of temperature on microbial growth has been widely studied, role proteome allocation in bringing about temperature-induced changes remains elusive. To tackle this problem, we propose a coarse-grained model growth, including processes temperature-sensitive protein unfolding and chaperone-assisted (re)folding. We determine sector that maximizes balanced rate as function nutrient limitation temperature. Calibrated with quantitative proteomic data for Escherichia coli , allows us to clarify general principles temperature-dependent formulate generalized laws. The same activation energy metabolic enzymes ribosomes leads an Arrhenius increase at constant composition over large range temperatures, whereas extreme temperatures resources are diverted away from chaperone-mediated stress responses. Our approach points risks possible remedies use ribosome content characterize complex ecosystems variation.

Temperature dependence of metabolic rates for microbial growth, maintenance, and survival.

Our work was motivated by discoveries of prokaryotic communities that survive with little nutrient in ice and permafrost, with implications for past or present microbial life in Martian permafrost and Europan ice. We compared the temperature dependence of metabolic rates of microbial communities in permafrost, ice, snow, clouds, oceans, lakes, marine and freshwater sediments, and subsurface aqu...

Analysis of the microbial proteome.

Proteomics has begun to provide insight into the biology of microorganisms. The combination of proteomics with genetics, molecular biology, protein biochemistry and biophysics is particularly powerful, resulting in novel methods to analyse complex protein mixtures. Emerging proteomic technologies promise to increase the throughput of protein identifications from complex mixtures and allow for t...
